Transplant and Autoimmune Therapy Using T-Cells Expressing Programmed Death Ligand-1 (PD-L1)
Programmed death (PD) ligand 1 (PD-L1) is an immune molecule present on regulatory T cells (Tregs), other suppressor cell populations, and tumor cells; the function of PD-L1 is to suppress the function of pathogenic T cells that express the PD1 receptor. Therefore, it has been hypothesized that the transfer of T cells that are enriched for PD-L1 expression might represent an effective method to suppress autoimmunity or transplant complications. Adoptive T cell therapy using Tregs is one such approach; however, this approach is limited due to the relative rarity of Tregs and their tendency to possess differentiation plasticity towards pathogenic T cell subsets such as the Th17 subset. Ex vivo co-stimulated and expanded effector T cells can be generated in sufficient numbers for cell therapy; however, such cells are not enriched for PD-L1 expression.
The current technology overcomes these limitations through transduction of co-stimulated T cells with a lentiviral expression vector that dictates T cell expression of PD-L1. In this method, the co-stimulated T cells acquire the immunosuppressant characteristics of Treg cells. The PD-L1 gene expression construct co-expresses a cell surface molecule (i.e., CD19 or CD34) that allows enrichment of the gene-modified T cells to high purity. Also the construct co-expresses another gene, TMPK, which acts as a safety cell fate switch because the TMPK can specifically activate the cytotoxic prodrug, AZT. By incorporation of this TMPK/AZT cell fate safety switch, the current technology will allow for PD-L1 therapeutic delivery, with subsequent elimination of the therapeutic cells in the event of toxicity. Statement by the Press Secretary on Key Trade Measures in H.R. 5986
Today, the President signed into law H.R. 5986, which includes key measures to bolster domestic manufacturing and strengthen our economic ties with sub-Saharan Africa and Central America. This bill also contains provisions to renew authorities under the Burma Freedom and Democracy Act. Congress passed this legislation with overwhelming bipartisan support.
This bill addresses sub-Saharan Africa’s top trade priority with the United States by extending a key provision under the African Growth and Opportunity Act (AGOA) trade preference program until September 2015. This provision, which was set to expire in the fall, will continue to allow duty-free access to the U.S. market for certain AGOA apparel exports. AGOA continues to be critical to expanding and diversifying our trade and investment relationship with Africa.
This legislation also includes technical amendments to textile and apparel provisions under the Central America – Dominican Republic – United States Free Trade Agreement (CAFTA-DR). These technical changes will have real job-supporting economic benefits for thousands of American workers in our textile and apparel industry, and will help boost economic growth in these developing regions.

Personalized Body Weight Management System Using Monitoring Devices and Mathematical Models of Metabolism
Scientists at the NIH have developed new methods for prescribing and monitoring personalized weight management interventions. The system uses validated mathematical models of human metabolism to set weight management goals and predict individual body weight outcomes in the context of changing metabolic needs and calorie consumption. The system uses repeated monitoring of a patient's body weight to assess progress and provide specific feedback to the patient and health care professional. Projected outcomes and body weight goals can be revised over time along with required prescription modifications to meet the body weight goals. The system is integrated into a network of one or more devices that may additionally monitor various physiological parameters, physical activities, food intake, or other behaviors. Such an enhanced personalized weight management program has great promise in the management of obesity. Kirsten Dunst Drinks Beer and Cheers on Team USA
Kirsten Dunst is the latest celebrity to get in on the Olympic action in London. Last night she drank beer and cheered from the crowd while Team USA beat Australia 119-86 in the women's basketball quarterfinals. Kristen was joined by a few friends, including the very handsome Oscar Isaac. Just last month, Kirsten was with her boyfriend, Garrett Hedlund, in NYC. Garrett and Kirsten met while filming On the Road and their big-screen chemistry can be seen in the latest On the Road trailer. The film opens in the UK Oct. 12, with the London premiere scheduled for next Thursday, but there's still no official release date for the States. There is, however, another important release date on the horizon for Kirsten. She shares the big screen with Isla Fisher, Lizzy Caplan, and James Marsden in the sure-to-be-hilarious comedy Bachelorette, which hits US theaters Sept. 7 in limited release.

Sofia Vergara Tells Allure - "If You Don’t' Have a Big A*s, You're Nothing!"

<P><a href="http://www.mtv.com/music/artist/bieber_justin/artist.jhtml">Justin Bieber</a> tweeted that when he was looking for inspiration for his <a href="/news/articles/1690963/justin-bieber-as-long-as-you-love-me-music-video-premiere.jhtml">"As Long As You Love Me" video</a>, there was no one he wanted to <a href="/news/articles/1691049/justin-bieber-as-long-as-you-love-me-michael-jackson.jhtml">emulate more than Michael Jackson</a>.</P><P>Coincidentally, when he needed someone to play his foe in the clip, he looked no further than a guy who has some experience working with Jackson.</P><P>That man is Michael Madsen, who appeared in Jackson's 2001 gangster-themed video for "You Rock My World" off his <i>Invincible</i> album. Oddly enough, the only people that have ever really asked the hard-edged actor to appear in videos happen to be Bieber and Jackson.</P><P></p><div class="player-placeholder right" title="Michael Madsen Has A Soft Spot For Justin Bieber" id="vid:822276" width="415" height="255"></div><p></P><P>"Hardly ever [am I asked to be in videos], that was why it was kind of funny cause that doesn't usually happen," he told MTV News. "It came out of nowhere. It was the last thing in the world I expected, to be called by Justin Bieber. It was kind of funny. It took me off guard. It's not the usual thing people think of me for, but considering the role, maybe."</P><P>In <a href="/news/articles/1690932/justin-bieber-as-long-as-you-love-me-video.jhtml">the video</a>, Madsen plays a tough guy who <a href="/news/articles/1691089/justin-bieber-michael-madsen-as-long-as-you-love-me-punch.jhtml">beats up Bieber</a> after the teen ignores his request to stay away from his daughter. It was that cinematic storyline that got Madsen interested.</P><P>"I didn't want to imagine myself on a trampoline with a lollipop, so I asked for the story. I realized it was almost like a film production; and of course, I kind of figured that it might be an interesting thing to do," he said about the Anthony Mandler-directed video. "I didn't know what to expect... I was surprised... They got me a good wardrobe. That was my shirt though. I brought my own shirt; those kinds of shirts are kind of rare.</P><P>"The story is so much Shakespearian; the couple, they run off together out the window," he later continued. "It's rather like that, I thought that was interesting. And I thought the kid, he had an enormous talent and he was very gracious to my sons and was very gracious to my family."</P><P>It was actually those sons that convinced the Hawaiian-shirt-wearing "Kill Bill" star to make the video with Bieber. "Well I was surprised at first that the kid was interested in having me be part of his video," he recalled. "I went to my sons and I asked my boys, I asked my sons what they thought if I should if they thought I should do it or not. And, of course, they gently placed their hand on my shoulder and said 'Dad don't you realize who Justin Bieber is?' And I said 'Well, no, actually I didn't.' Coldplay, Led Zeppelin Album Covers Featured On New British Stamps

<P>On Thursday (January 7), the U.K.'s Royal Mail unveiled a series of 10 new stamps, honoring the most iconic album covers of the past 40 years.</P><P>The albums featured in the new series are the <MTVNLINK type="artist" id="515">Rolling Stones</MTVNLINK>' <i>Let It Bleed,</i> <MTVNLINK type="artist" id="993">Led Zeppelin</MTVNLINK>'s <i>IV,</i> <MTVNLINK type="artist" id="973">David Bowie</MTVNLINK>'s <i>The Rise and Fall of Ziggy Stardust and the Spiders From Mars,</i> <MTVNLINK type="artist" id="503245">Mike Oldfield</MTVNLINK>'s <i>Tubular Bells,</i> <MTVNLINK type="artist" id="977">The Clash</MTVNLINK>'s <i>London Calling,</i> <MTVNLINK type="artist" id="267">New Order</MTVNLINK>'s <i>Power, Corruption and Lies,</i> <MTVNLINK type="artist" id="758">Primal Scream</MTVNLINK>'s <i>Screamadelica,</i> <MTVNLINK type="artist" id="1118">Pink Floyd</MTVNLINK>'s <i>The Division Bell,</i> <MTVNLINK type="artist" id="1041">Blur</MTVNLINK>'s <i>Parklife</i> and — the most recent album on to make the cut — <MTVNLINK type="artist" id="1111141">Coldplay</MTVNLINK>'s <i>A Rush of Blood to the Head.</i></P><P>Meant to honor "the most potent graphic images of modern times, many of which have provided a visual soundtrack to people's lives," the series is the end result of a lengthy research process by the Royal Mail, who looked through thousands of album covers by British artists before deciding on the final list. And, during a Wednesday night BBC Radio broadcast, it was revealed that the queen herself actually approved each design.</P><P>Led Zeppelin guitarist Jimmy Page — who helped design the cover for <i>IV</i> — was on hand to celebrate the release of the stamps and recalled the mysterious nature of the album's iconic imagery.</P><P>"Almost 40 years after the album came out, nobody knows the old man who featured on the cover, nor the artist who painted him," he said. "That sort of sums up what we wanted to achieve with the album cover, which has remained both anonymous and enigmatic at the same time."</P><P>Of course, any great honor is befitting of an equally great contest, so, on the same day the stamps were made available to the general public, Coldplay decided to give one of their <i>Rush of Blood</i> stamps away. In a message on their official site, the band held a contest to send one lucky fan "a letter using a Coldplay stamp, postmarked with today's issue date."</P><P>"We visited our local post office earlier today and bought some of the Coldplay stamps," the message read. "Very nice they are too."</P>

"We are trying out a couple of singers," he said. "We want to do it. It's sounding great, and we want to get on and get out there."</P><P>But Jones insists that he, Page and Bonham aren't interested in finding a Plant clone. "It's got to be right," he said. "There's no point in just finding another Robert. You could get that out of a tribute band, but we don't want to be our own tribute band. There would be a record and a tour, but everyone has to be onboard."</P><P>Of course, even if Zeppelin were to approach him, Cornell might be too busy to take over vocals for the band. His upcoming <a href="http://www.mtv.com/music/artist/timbaland/artist.jhtml">Timbaland</a>Timbaland-produced solo LP, <a href="/news/articles/1596730/chris-cornell-debuts-timbaland-produced-track-on-tv-show.jhtml"><i>Scream,</i></a> will be released within the next few months. Cornell told MTV News last month that some of his fans might consider <i>Scream</i> something of a departure, but he doesn't see it that way.</P><P>"It makes me happy that there's this perception that I have a group of fans that I'm now sort of throwing a curve at and what their reaction will be," he said. "But I've been in this situation so many times already. When I put out [1999's] <i>Euphoria Morning,</i> my main goal was to create an album that sounded like nothing I'd done in Soundgarden, and I did that. I also had that with Temple of the Dog, where I showed up with songs that weren't necessarily riff-based. And then, of course, the pairing of me and members of <a href="http://www.mtv.com/music/artist/rage_against_the_machine/artist.jhtml">Rage Against the Machine</a>Rage Against the Machine had everyone sort of speculating about what that would sound like.</P><P>"It feels like I've done this so many times that when it's presented to me as being a departure, I feel that's a misconception," he added. "I feel like that's my theme at this point."</P>

Robert Plant And Alison Krauss Win Grammy Album Of The Year
Raising Sand, from Led Zeppelin vet and bluegrass superstar, wins five Grammys on Sunday night.

<P>"I'm bewildered," <MTVNLINK type="artist" id="14245">Robert Plant</MTVNLINK> said onstage as he accepted the <MTVNLINK type="news" id="1604580">Grammy Album of the Year award</MTVNLINK> with <MTVNLINK type="artist" id="506074">Alison Krauss</MTVNLINK> on Sunday night. "In the old days we would have called this selling out, but it's a good way to spend a Sunday."</P><P>He was probably one of the few who were surprised, because <i>Raising Sand,</i> which won five trophies at Sunday night's show, is in many ways the perfect <a href="http://www.mtv.com/news/grammys/">Grammy</a> album. It features two respected veterans, a critically lauded producer, some sandpaper-and-velvet vocals and a baker's dozen of time-tested standards.</p><div style="margin: 0px 10px 10px 0px; float: left;"><embed src="http://media.mtvnservices.com/mgid:uma:video:mtv.com:338316" width="256" height="223" type="application/x-shockwave-flash" flashVars="configParams=instance%3Dnews%26vid%3D338316" allowFullScreen="true" allowScriptAccess="always" base="."></embed></div><p></P><P>You're probably familiar with Robert Plant from his <MTVNLINK type="artist" id="993">Led Zeppelin</MTVNLINK> days, and you might be aware of producer T-Bone Burnett's work on the "O Brother, Where Art Thou?" soundtrack (it won the Grammy for Album of the Year in 2002). And if you don't know who Alison Krauss is, she possesses a haunting set of pipes and is one of the meanest fiddle players in the world. Oh, and she's won 21 Grammys, more than any other female artist and the seventh-most in history.</P><P>Really, she's the key to <i>Sand</i>'s success, and not just because of her voice (or her fiddle playing). She and Plant first met in 2004, at a Rock and Roll Hall of Fame tribute to legendary bluesman Leadbelly, and the former Zeppelin man was amazed by her knowledge of American Roots music — so much so that they began kicking around the idea of recording an album together. Three years later, <i>Sand</i> was released.</P><P>And while Plant possesses the more famous voice, the album's finest moments radiate from Krauss. Whether she's getting bluesy on Little Milton's "Let Your Loss Be Your Lesson" or entwining with Plant's husky voice on songs like "Please Read the Letter" and Roly Salley's winsome "Killing the Blues," she more than carries her end of the bargain.</P><P>And perhaps that's also due to producer Burnett, who handpicked the 13 songs the duo cover on <i>Sand.</i> His arrangements are sparse — giving the two voices ample room to breathe — yet dense, warm and crackling at the same time. It's a testament to his work that he's often given just as much billing as Plant and Krauss on the project ... and it's certainly justified.</P><P>To date, <i>Sand</i> has sold more than 1 million copies, heaped tons of acclaim and actually earned a Grammy last year — "Gone Gone Gone (Done Moved On)" took home the award for Best Pop Collaboration With Vocals.</P><P>One expert was surprised not by the album's success, but by the fact that it's actually quite a good album.</P><P>"At first, the album seemed like a vanity project. ... Two names, clearly a one-off record, didn't have to be any good, you know?" <i>New York Times</i> music critic Jon Caramanica said. "Led Zeppelin fans would buy it because of Robert Plant, Alison Krauss would get a check. But it actually turned out to be a really thoughtful, really <i>good</i> record. So when you combine all that with the fact that the Grammys love to lionize one of their own, I could really see it taking home some awards." </P>

And if fans were wondering what and who influenced the video, Bieber took to <a href="https://twitter.com/justinbieber" target="_blank">Twitter</a> hours after the premiere to shout out his hero, Michael Jackson, for helping him step up his music-video game.</P><P>"MJ told stories with his videos. I wanted to tell a story. Feel like a movie. #AsLongAsYouLoveMe," he wrote, showing some love to the master. He later added, "Push the limits. Fight for what you BELIEVE in."</P><P>As of Wednesday night, Bieber was astounded that his video had amassed more than 1 million views in less than 10 hours. But that number nearly quadrupled on Thursday morning — as of press time, the video had almost 4 million streams.</P><P>Bieber is not only feeling the love for his <a href="/news/articles/1690932/justin-bieber-as-long-as-you-love-me-video.jhtml">"ALAYM" video</a> but also for his "Boyfriend" video, which just received two <a href="http://www.mtv.com/ontv/vma/2012/">2012 MTV Video Music Award</a> nominations for <a href="/news/articles/1690978/rihanna-only-girl-best-pop-video-video-music-awards.jhtml">Best Pop Video</a> and Best Male Video. Bieber goes into the September 6 broadcast already a two-time VMA winner. He'll learn if he can take home another one when the show goes live from the Staples Center.</P><P>The VMAs take place just weeks before Bieber kicks off his sold-out <a href="/news/articles/1688265/justin-bieber-after-believe.jhtml"><i>Believe</i> tour</a>, which coincidentally will also be heavily influenced by the King of Pop.</P><P>"He has been watching a tremendous amount of Michael Jackson concerts from the '80s and '90s," his manager Scooter Braun shared. "And he has been talking to me about how Michael incorporated magic and pyro and reveals and he wants to bring in that. [But] still do that acoustic thing, but he wants to take it to a whole new level.</P><P>"It's just really exciting," Braun continued. "Nothing makes him happier than being onstage. So it's gonna be fun to get back onto the road. I think he was an amazing performer before, but his level of dance and musicianship and his range [has grown]. He's a veteran now. I think he wants to take it to a completely new level."</P>
